US Highway 341's Southern Terminus is in Brunswick at US Highway 17 just north of The Sidney Lanier Bridge. From there it follows Fourth Ave, Newcastle St and Prince St through
a mix of residential and industrial areas before turning onto Bay St with the Georgia Ports Authority's Mayors Point Docks to the west and
The Downtown Brunswick Central Business District on the east, joining with US25 along the way. After rejoining Newcastle St just north of Downtown Brunswick, US341/25
continues north as Newcastle Street Extension, Norwich Street Extension and New Jesup Highway as it proceeds through mostly commercial areas through Dock Junction and past
Interstate Highway 95's Exit 36 interchange. US341/25 then becomes semi rural through Sterling, GA and then mostly rural from Sterling to the
Wayne County Line. From there, US341 and US25 will continue on to Jesup, GA where the two highways part ways with US341 continuing on to its Northern terminus at US41 in Perry, GA and
US25 continuing on to its Northern terminus at US42/127 in Cincinnati, OH.
